1. Carbachol induced Ca oscillation in SH-SY5Y cell line.
2. Stimulation-induced [Ca2+]i Oscillation Patterns
3. Effects of [Ca2+]o on the Oscillation
4. Effect of Ca channel antagonist's effects on the oscillations
5. Effects of the carbachol's concentration on the oscillation
( Figure not given)The oscillation frequency change wherease the peak amplitude stay unchanged.
6. Possible Mechanism
Muscarinic receptor agonist ---carbachol , can activated PLC . Then PLC can accelerate the PIP2 hydrolysis process. When the InsP3 has been accumulated to a level, it can lead to the calcium release from the InsP3-type calcium store.
And the calcium entering the cytosolic can also activated PLC and produced a positive feedback, so form the quick uprising phase of the peak. Then high [Ca2+]i will activate Ca2+ -ATPase pump both on the membrane of the cell and the ER, and pump Ca2+ back to the store or out of the cell, as the same time muscarine may open some calcium entry for a short time , and form the plateau phase which represent a balance among the three factors' effects mentioned above.
When the [Ca2+]i come back to the base line, and calcium store has been replenish to a certain extent, it can be triggered to release calcium for the second time, so come out the calcium oscillations process.
